Biological Background: CD271 Function and Localization
- CD271, also known as p75NTR, Gp80-LNGFR, or TNFRSF16, is a tumor necrosis factor receptor superfamily member that functions as a low-affinity receptor for neurotrophins including NGF, BDNF, NTF3, and NTF4.
- Forms a heterodimeric receptor with SORCS2 that binds precursor forms of NGF, BDNF, and NTF3 with high affinity, while showing much lower affinity for mature neurotrophins. Related references: PMID:24908487
- Plays a dual role in neuronal cell fate, capable of mediating both cell survival and apoptosis depending on context and co-receptor interactions.

- The immunogen corresponds to amino acids 330-427 of human CD271 (intracellular domain), which may provide specific detection of the full-length receptor. For IHC-P, consider using heat-induced antigen retrieval in citrate buffer (pH 6.0) and validate staining in positive control tissues such as human brain, nerves, or melanoma samples.
- For ELISA applications, this polyclonal antibody can be used as a capture or detection reagent. Optimize coating antigen (recombinant CD271 protein or cell lysates) and antibody titration to achieve optimal sensitivity. Cross-reactivity with related neurotrophin receptors should be assessed.
- As a rabbit polyclonal antibody, lot-to-lot consistency should be verified in the intended assay system. Include appropriate negative controls (e.g., tissue from CD271-knockout models or non-expressing cell lines) to confirm specificity.
