Biological Background: CD27 Function and Localization
- CD27 (TNFRSF7) is a single-pass type I membrane protein and member of the tumor necrosis factor receptor superfamily, functioning as a costimulatory immune-checkpoint receptor primarily expressed on T-cells, NK-cells, and B-cells. Related references: PMID:28011863
- Upon binding its ligand CD70/CD27L on B-cells, CD27 activates the TRAF2-PTPN6 axis, which inhibits LCK phosphorylation to drive phenotypic and transcriptional adaptations essential for T-cell memory formation (PubMed:38354704). Related references: PMID:38354704
- Beyond T-cell activation, CD27 engagement on early progenitor cells provides a negative feedback signal to leukocyte differentiation, thereby modulating hematopoiesis (By similarity), and negatively regulates Th2 lymphocyte function in adipose tissue (By similarity).
- The receptor is a glycoprotein and phosphoprotein with three cysteine-rich domains (disulfide bonds) and is localized to the cell membrane.
- It is broadly expressed in most T-lymphocytes and has been associated with apoptosis signaling and disease variants, including lymphoproliferative syndrome 2 (LPFS2).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 20-191) of human CD27, suggesting the antibody recognizes the ligand-binding region; consider this when designing competitive assays or epitope mapping studies.
- For Western blotting, the predicted molecular weight is 29 kDa; however, glycosylation may cause a shift to higher apparent molecular weights – validate in your sample system with appropriate controls.
- For immunohistochemistry on paraffin-embedded sections (IHC-P), antigen retrieval using heat-induced epitope retrieval (HIER) in citrate buffer (pH 6.0) is recommended; optimize incubation times and antibody dilution for best signal-to-noise.
- This polyclonal antibody reacts with human, mouse, and rat CD27, enabling cross-species studies; however, confirm reactivity in your specific model before critical experiments.
- Multiple applications are reported (WB, IF/ICC, IF-P, IHC-P, ELISA); titrate the antibody for your specific assay format and include appropriate positive and negative controls.
