Biological Background: CD27 Function and Localization

  • CD27 (TNFRSF7) is a single-pass type I membrane protein and member of the tumor necrosis factor receptor superfamily, functioning as a costimulatory immune-checkpoint receptor primarily expressed on T-cells, NK-cells, and B-cells. Related references: PMID:28011863
  • Upon binding its ligand CD70/CD27L on B-cells, CD27 activates the TRAF2-PTPN6 axis, which inhibits LCK phosphorylation to drive phenotypic and transcriptional adaptations essential for T-cell memory formation (PubMed:38354704). Related references: PMID:38354704
  • Beyond T-cell activation, CD27 engagement on early progenitor cells provides a negative feedback signal to leukocyte differentiation, thereby modulating hematopoiesis (By similarity), and negatively regulates Th2 lymphocyte function in adipose tissue (By similarity).
