Biological Background: CD24 Function and Localization

  • CD24 is a signal transducer protein, also known as small cell lung carcinoma cluster 4 antigen, encoded by the CD24 (CD24A) gene. It is a GPI-anchored glycoprotein with a molecular weight of about 8 kDa.
  • CD24 plays a pivotal role in cell differentiation across different cell types. Signaling may be triggered by lectin-like ligand binding to CD24 carbohydrates, leading to second messenger release from the GPI anchor.
  • In B-cells, CD24 modulates activation responses. It promotes antigen-dependent proliferation of B-cells and prevents their terminal differentiation into antibody-forming cells. Related references: PMID:11313396
  • In association with SIGLEC10, CD24 may help selectively suppress immune responses to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90.
  • CD24 contributes to the control of autoimmunity (by similarity).
  • The protein localizes to the cell membrane via its GPI anchor.
  • CD24 is expressed predominantly on B-cells, certain T-cells, and various cancer cell lines including erythroleukemia and small cell lung carcinoma.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 1-80) covers the N-terminal extracellular region of human and mouse CD24. This region is expected to be exposed on the cell surface in native conditions, supporting applications such as IF/ICC and IF-P.
  • In WB, the predicted molecular weight is around 8 kDa for the unglycosylated human protein, though observed bands may appear larger or display multiple forms due to glycosylation. The murine homolog may run at approximately 13 kDa. Consider using low-molecular-weight markers and optimized transfer conditions for small proteins.
  • For IF/ICC and IF-P, expect membrane staining. Validate fixation and permeabilization conditions, as the GPI anchor may be sensitive. Use appropriate controls to confirm specificity.
  • In ELISA, the antibody may be used for capture or detection of recombinant or native CD24. Titration is recommended to determine the optimal working concentration.
