Biological Background: CD274 Function and Localization
- CD274 (PD-L1/B7-H1) is a type I transmembrane protein of the immunoglobulin superfamily, encoded by the Cd274 gene in mouse (aliases: B7h1, Pdcd1l1, Pdcd1lg1, Pdl1).
- Acts as a ligand for the inhibitory receptor PD-1 (PDCD1), playing a critical role in maintaining immune tolerance by modulating T-cell activation and limiting effector responses (PubMed:11238124). Related references: PMID:11238124
- May co-stimulate T-cell subsets that primarily produce interleukin-10 (IL10) through a yet unidentified activating receptor (PubMed:11015443, PubMed:12719480). Related references: PMID:11015443, PMID:12719480
- The PDCD1/PD-L1 pathway is exploited by tumors to attenuate anti-tumor immunity; interaction with PD-1 inhibits cytotoxic T lymphocyte (CTL) effector function, and blockade of this pathway reverses T-cell exhaustion, providing a rationale for cancer immunotherapy (PubMed:12218188, PubMed:27281199). Related references: PMID:12218188, PMID:27281199
- Subcellular localization includes the cell membrane, early endosome membrane, and recycling endosome membrane.
- Highly expressed in the heart, thymus, skeletal muscle, and lung; weakly in the kidney, spleen, thyroid, and liver. Expressed on activated dendritic cells, B-cells, macrophages, and numerous lymphoid tumor cell lines.
- Structural features include immunoglobulin-like domains, a disulfide bond, and glycosylation; also possesses a signal peptide, a transmembrane helix, and ubiquitination sites (UniProt keywords).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 19-239) of mouse CD274, covering the region responsible for PD-1 interaction; this design may favor detection of surface-expressed and soluble forms of the protein in mouse samples.
- For Western blotting, consider using reducing SDS-PAGE conditions; the predicted unmodified molecular weight is ~33 kDa, but glycosylation often results in a higher apparent molecular weight (typically 40–50 kDa). Include appropriate controls to confirm band identity.
- When performing ELISA, titrate the antibody to determine optimal working concentrations; validate performance using recombinant CD274 protein or CD274-expressing cells to establish sensitivity and specificity.
- Although cross-reactivity with human and rat is indicated, verify binding characteristics in your specific experimental context, as epitope accessibility may vary across species and sample types.
