Biological Background: CD117 Function and Localization

  • CD117, also known as c-Kit or mast/stem cell growth factor receptor Kit, is a receptor tyrosine kinase encoded by the Kit proto-oncogene (SWISS: P05532). It belongs to the type III receptor tyrosine kinase family featuring extracellular immunoglobulin-like domains and an intracellular kinase domain.
  • Upon binding its ligand KITLG/SCF, CD117 dimerizes and autophosphorylates, triggering downstream signaling pathways including PI3K-AKT, RAS-MAPK/ERK, and JAK-STAT. These cascades regulate cell survival, proliferation, hematopoiesis, stem cell maintenance, gametogenesis, melanogenesis, and mast cell development.
  • Multiple isoforms exist due to alternative splicing. Isoform 1 and isoform 2 are found in bone marrow and spermatogonia; isoform 3 is enriched in hematopoietic stem cells and detected in later stages of spermatogenesis.
