Biological Background: CD27 Function and Localization
- CD27 (TNFRSF7/T14) is a costimulatory immune-checkpoint receptor expressed on T-cells, NK-cells, and B-cells, where it binds to and is activated by its ligand CD70/CD27L on B-cells. Related references: PMID:28011863
- The CD70-CD27 signaling pathway mediates antigen-specific T-cell activation and expansion, which in turn provides immune surveillance of B-cells. Related references: PMID:28011863
- Upon CD70 ligation, CD27 recruits TRAF2 and activates PTPN6, inhibiting LCK phosphorylation and driving transcriptional adaptations essential for T-cell memory formation. Related references: PMID:38354704
- The protein is localized to the cell membrane and is found in most T-lymphocytes.
- Post-translational modifications include disulfide bond formation, glycosylation, and phosphorylation, as annotated in UniProt.
- CD27 also delivers a negative feedback signal to early hematopoietic progenitor cells, thereby modulating leukocyte differentiation during immune activation.
- The mature protein has a calculated molecular weight of approximately 29 kDa (UniProt: 29137 Da).
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 21–192 of human CD27, which covers the extracellular ligand-binding domain. Consequently, the antibody is expected to recognize the native CD27 receptor on live cells, making it suitable for flow cytometry (FC) applications.
- For flow cytometry, consider using appropriate Fc receptor blocking reagents to minimize non-specific binding, especially when analyzing lymphocyte populations rich in Fc receptor-expressing cells.
- Validate the APC conjugate in your specific sample type and instrument setup; fluorophore conjugation may subtly alter antibody affinity or background staining.
