Biological Background: Syntaxin-1A Function and Localization
- Syntaxin-1A (STX1A), also known as HPC-1, is a Q-SNARE protein and a key member of the syntaxin family.
- Essential for calcium-dependent exocytosis of neurotransmitters and hormones; forms the SNARE complex with SNAP25 and VAMP2 to drive synaptic vesicle fusion with the plasma membrane (PubMed:26635000). Related references: PMID:26635000
- Participates in the calcium-dependent regulation of acrosomal exocytosis in sperm, highlighting its role beyond neuronal tissues (PubMed:23091057). Related references: PMID:23091057
- Involved in the regulated secretion of insulin and glucagon-like peptide 1 (GLP-1), linking it to metabolic processes.

- The immunogen spans amino acids 1-288 of human STX1A, covering a large portion of the protein; thus, the polyclonal antibody may recognize multiple epitopes, potentially enhancing sensitivity in applications such as Western blotting.
- For Western blot (WB), a band at approximately 33 kDa is expected; use whole-cell or tissue lysates from human or rat samples, and include appropriate loading controls and molecular weight markers.
- In immunofluorescence/immunocytochemistry (IF/ICC), optimal fixation and permeabilization conditions should be determined empirically, as STX1A localizes to the plasma membrane and intracellular vesicles.
- For ELISA, the recombinant immunogen can be used as a positive control to establish assay parameters.
- Given reported cross-reactivity with rat, validation in rat-derived tissues or cell lines is feasible; however, cross-reactivity with other species should be independently tested if required.
