Biological Background: PKC zeta Function and Localization
- PKC zeta (nPKC-zeta) is an atypical protein kinase C, lacking calcium and diacylglycerol sensitivity, and functions as a serine/threonine-protein kinase in NF-kappa-B activation, mitogenic signaling, cell proliferation, cell polarity, inflammatory response, and maintenance of long-term potentiation (LTP).
- Downstream of PI3K, PKC zeta activates the MEK1-ERK2 cascade independently of RAF1 in macrophages and following mitogenic stimuli. It also contributes to insulin-dependent AKT3 activation and GLUT4 translocation in adipocytes, acting partly as an adapter.
- PKC zeta mediates NF-kappa-B activation via interactions with SQSTM1/p62, RIPK1, and TRAF6, leading to IKBKB phosphorylation and NF-kappa-B inhibitor degradation. It further phosphorylates RELA at Ser-311 to counteract SETD6-mediated repression.
- In cell polarity, PKC zeta forms a cytoplasmic complex with PARD6A and is recruited by CDC42 to establish polarity with microtubule motors. It stimulates neuronal differentiation with FEZ1 and is required for late-phase LTP in hippocampal CA1 synapses, crucial for long-term memory.
- PKC zeta phosphorylates VAMP2 in vitro and activates LRRK1, which then phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:17313651 PMID:36040231
- PKC zeta localizes to the cytoplasm, endosomes, cell junctions, and membrane. It is predominantly expressed in the brain, with lower levels in lung, kidney, and testis. It contains a zinc-finger domain and ATP-binding site, and is subject to alternative splicing and phosphorylation.

- The immunogen corresponds to the C-terminal region (amino acids 476–547), which may provide good epitope accessibility for native and denatured protein detection.
- For Western blotting, the predicted molecular weight is ~68 kDa; always include positive and negative controls to confirm specificity.
- In immunofluorescence/ICC, optimize fixation and permeabilization conditions, as PKC zeta can localize to multiple compartments (cytoplasm, membrane, endosomes, junctions).
- ELISA can be used for quantitative detection of PKC zeta in cell lysates or purified protein; validate with known standards.
