Biological Background: p63 Function and Localization

• Tumor protein 63 (p63), also known as p73L, p40, p51, and KET, is a member of the p53 family of transcription factors. Multiple isoforms are generated through alternative promoter usage and alternative splicing, each containing varying transactivation and inhibitory domains, resulting in isoform-specific activities.
• Acts as a sequence-specific DNA binding transcriptional activator or repressor, regulating genes involved in development and cellular stress responses.
• Plays a role in Notch signaling by probably inducing expression of JAG1 and JAG2, contributing to cell fate decisions.
• Involved in the regulation of epithelial morphogenesis; the ratio of DeltaN-type and TA*-type isoforms governs epithelial stem cell maintenance and the initiation of epithelial stratification.
• Required for limb formation from the apical ectodermal ridge during embryonic development.

• For Western blot (WB), anticipate bands within the 45-77 kDa range, consistent with the reported molecular weight of different p63 isoforms. Optimize gel percentage and transfer conditions for this size range.
