Biological Background: eIF4E Function and Localization

Eukaryotic translation initiation factor 4E (eIF4E), also known as eIF4E1, eIF-4F 25 kDa subunit, or mRNA cap-binding protein, is encoded by the EIF4E gene (Gene ID: 1977). It serves dual functions in the cytoplasm for translation initiation and in the nucleus for mRNA export (PubMed:11606200, PubMed:22578813, PubMed:22684010).

eIF4E specifically recognizes and binds the 7-methylguanosine (m7G) cap at the 5' end of mRNAs, a critical step that facilitates ribosome recruitment and unwinding of mRNA secondary structures (PubMed:16271312, PubMed:22578813). Together with EIF4G1, it enables TISU element-dependent translation, bypassing the need for ribosomal scanning (PubMed:29987188).
