Biological Background: BSN (Bassoon) Function and Localization
- BSN encodes protein bassoon (also known as zinc finger protein 231), a large scaffold protein of the presynaptic cytomatrix at the active zone (CAZ) where neurotransmitter is released (PubMed:12812759).
Related references: PMID:12812759 - After synthesis, bassoon participates in the formation of Golgi-derived membranous organelles termed Piccolo-Bassoon transport vesicles (PTVs) that are transported along axons to sites of nascent synaptic contacts (PubMed:19380881).
Related references: PMID:19380881 - At the presynaptic active zone, bassoon regulates the spatial organization of synaptic vesicle clusters, the protein complexes that execute membrane fusion, and compensatory endocytosis.
- Bassoon also plays roles in regulating presynaptic protein ubiquitination by interacting with SIAH1 and in regulating presynaptic autophagy by associating with ATG5.
- Bassoon mediates synapse-to-nucleus communication by associating with the transcriptional corepressor CTBP1, reducing the size of its pool available for nuclear import and thereby reconfiguring gene expression.
- The protein inhibits the activity of the proportion of DAO enzyme that localizes to the presynaptic active zone, which may modulate synaptic transmission.
- BSN is exclusively expressed in brain tissue, consistent with its critical synaptic functions.
Experimental Guidance and Technical Tips
- Due to the large molecular weight of bassoon (approximately 416 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6% or gradient gels) and extended transfer times for western blotting.
- For immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization protocols to preserve synaptic structures; bassoon localizes to presynaptic active zones, so co-staining with synaptic markers is recommended.
- The immunogen corresponds to amino acids 830-950 of human BSN; verify antibody specificity in relevant knockout or knockdown models.
- This antibody shows cross-reactivity with mouse BSN, making it suitable for translational studies in mouse brain tissue.
- For ELISA applications, validate the antibody pair compatibility and assay conditions with purified recombinant protein or native tissue lysates.
