IL-6 (Interleukin-6)

Target Introduction:

IL-6 (Interleukin-6) is a pleiotropic cytokine that plays a crucial role in the immune system and other biological systems. It participates in various physiological events in different organs, including inflammation, hematopoiesis, and oncogenesis. IL-6 regulates cell growth, gene activation, proliferation, survival, and differentiation.

IL-6 signals through a receptor complex composed of two subunits: an alpha subunit (responsible for ligand specificity) and GP130 (Glycoprotein 130), a receptor subunit shared by other cytokines in the IL-6 family. Binding of IL-6 to its receptor initiates cellular events such as:

  • Activation of JAK (Janus Kinase) kinases
  • Activation of Ras-mediated signaling.

Activated JAK kinases phosphorylate and activate STAT transcription factors, particularly STAT3 (Signal Transducers and Activators of Transcription-3) and SHP2 [SH2 (Src Homology-2) Domain-containing Tyrosine Phosphatase]. Phosphorylated STAT3 dimerizes, translocates to the nucleus, and activates transcription of genes containing STAT3 response elements. STAT3 is crucial for GP130-mediated cell survival and G1 to S cell-cycle-transition signals. c-Myc and Pim are identified as STAT3 target genes, compensating for STAT3 in cell survival and cell-cycle transition. SHP2 links the cytokine receptor to the Ras/MAP (Mitogen-Activated Protein) kinase pathway and is essential for mitogenic activity.

Applications in IVD:

IL-6 is used as a target in In Vitro Diagnostics (IVD). Reportedly, it can be used in LF (Lateral Flow Assay) and FIA (Fluorescence Immunoassay).
