- Polycomb protein EED (Embryonic ectoderm development protein), also designated HEED, COGIS, and WAIT-1, is a core component of the polycomb repressive complex 2 (PRC2). Alternative splicing and alternative initiation yield multiple isoforms.
- As part of the PRC2/EED-EZH2 complex, EED methylates histone H3 at lysine-9 and lysine-27, leading to transcriptional silencing of target genes such as HOXC8, HOXA9, MYT1, and CDKN2A.
- EED recognizes trimethylated histone H1 at lysine-26 (H1K26me3), which inhibits PRC2 methyltransferase activity on nucleosomal H3K27, while recognition of H3K27me3 propagates the repressive mark.
- The PRC2/EED-EZH2 complex may also recruit DNA methyltransferases, establishing a functional link between histone modification and DNA methylation for stable gene repression.
- EED is localized to the nucleus and associates with chromosomes, consistent with its role in chromatin regulation.

Experimental Guidance and Technical Tips
- The immunogen encompasses the N-terminal 400 amino acids of human EED; this region may detect full-length protein and certain isoforms. Validate detection of any specific splice variants in your experimental system.
- For Western blotting (WB), the predicted molecular weight is approximately 50 kDa. Confirm band size with appropriate positive and negative controls.
- Cross-reactivity is reported for human, mouse, and rat. Verify reactivity with other species if needed by sequence alignment of the immunogen.
- In immunofluorescence/ICC, perform standard fixation (e.g., 4% PFA) and permeabilization (e.g., 0.1% Triton X-100). Nuclear staining is expected.
