Biological Background: TRMT2A Function and Localization
- TRMT2A (tRNA (uracil-5-)-methyltransferase homolog A), also known as HTF9C, is a member of the RNA methyltransferase family.
- It catalyzes the formation of 5-methyl-uridine (m5U) in cytoplasmic tRNAs, primarily at position 54 (m5U54), and in a subset of mRNAs, using S-adenosyl-L-methionine as the methyl donor. Related references: PMID:31361898 | PMID:33799331 | PMID:34556860 | PMID:34123281
- The protein contains a coiled-coil domain and undergoes alternative splicing and phosphorylation, contributing to functional diversity.
- It is predominantly localized in the cytoplasm/cytosol, consistent with its role in tRNA and mRNA processing.
- TRMT2A is classified under multiple functional keywords: methyltransferase, transferase, tRNA processing, mRNA processing, and RNA-binding.
- A 3D structure has been resolved, and its molecular weight is approximately 68.7 kDa, matching the observed 69 kDa band in experimental settings.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide from the C-terminal region (aa 500–end) of human TRMT2A. In western blot (WB), expect detection of a ~69 kDa band corresponding to full-length TRMT2A.
- For immunohistochemistry (IHC-P), consider optimizing antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 8.0) based on tissue type and fixation.
- In ELISA, the antibody can serve as a detection or capture reagent. Validate against recombinant TRMT2A or endogenous lysates from human, mouse, or rat samples.
- Cross-reactivity with mouse and rat has been confirmed; however, due to sequence homology, validate specificity in each species-specific application.
- Since this is a polyclonal antibody, lot-to-lot consistency should be verified when transitioning between production batches.
