Biological Background: TFAM Function and Localization
- TFAM (Transcription Factor A, Mitochondrial) is a member of the HMGB protein family and exists in several isoforms due to alternative splicing.
- Binds to the mitochondrial light strand promoter and functions as a key transcriptional activator. Related references: PMID:29445193 PMID:32183942
- Forms the mitochondrial transcription initiation complex with TFB2M and POLRMT; recruits POLRMT to promoters and induces structural changes enabling transcription. Related references: PMID:29149603 PMID:20410300
- Enhances mitochondrial RNA polymerase promoter recognition and promotes transcription initiation from HSP1 and light strand promoters; possesses DNA unwinding activity. Related references: PMID:22037172
- Bends the light strand promoter DNA into a U-turn shape via its HMG boxes, facilitating transcription initiation. Related references: PMID:1737790
- Required for maintenance of normal mitochondrial DNA levels; may also organize and compact mtDNA. Related references: PMID:19304746 PMID:22841477 PMID:22037171
- Localizes to the mitochondrial matrix and nucleoids; undergoes phosphorylation.
- Pathogenic mutations in TFAM cause mitochondrial DNA depletion syndrome 15 (MTDPS15), a primary mitochondrial disease.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 100-200 of human TFAM; this region may be conserved in mouse and rat, consistent with cross-reactivity data.
- For Western blot (WB), the predicted molecular weight is approximately 29 kDa; consider using whole-cell or mitochondrial extracts for optimal detection.
- For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), standard antigen retrieval and permeabilization protocols are recommended; validate in appropriate positive and negative control tissues.
- For ELISA, the antibody may be used as a capture or detection reagent; titrate to determine optimal working concentration.
- Always perform pilot experiments to confirm specificity and performance in your specific assay system and sample type.
