Biological Background: RNH1 Function and Localization

  • Ribonuclease inhibitor (RNH1), also known as placental ribonuclease inhibitor or RAI, is a ubiquitously expressed cytoplasmic protein that belongs to the leucine-rich repeat (LRR) family.
  • Functions as a potent inhibitor of pancreatic-type ribonucleases, including RNASE1, RNASE2, and angiogenin (ANG), through tight binding that blocks their catalytic activity. Related references: PMID:12578357 PMID:14515218 PMID:3219362
  • Plays a critical role in redox homeostasis by modulating intracellular RNase activity in response to oxidative stress. Related references: PMID:17292889
  • Constitutively inhibits the cytotoxic tRNA ribonuclease activity of ANG in the cytoplasm under non-stress conditions, preserving translational integrity. Related references: PMID:23843625 PMID:32510170
  • Upon cellular stress, RNH1 translocates to the nucleus, releasing cytoplasmic ANG inhibition while suppressing nuclear angiogenic activity of ANG, implicating RNH1 in stress-responsive signaling. Related references: PMID:23843625
  • Post-translational modifications include acetylation and phosphorylation, which may regulate its activity or subcellular localization; disease-associated variants have been reported, underscoring its physiological importance.
  • RNH1 contains multiple leucine-rich repeats that form a horseshoe-shaped structure, providing an extended binding interface for diverse RNases.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 202–461 of human RNH1 (NP_002930.2), encompassing critical LRR domains that are highly conserved across human, mouse, and rat; therefore, consider validating cross-reactivity in your specific sample system.
  • For Western blotting, a band of approximately 50 kDa is expected; include appropriate positive and negative controls and optimize primary antibody dilution.
  • For immunofluorescence/immunocytochemistry, fixation and permeabilization conditions may influence detection; we recommend testing both cytoplasmic and nuclear staining patterns, particularly under stress conditions where RNH1 relocates.
  • As an ELISA-suitable antibody, it may be used for quantification of RNH1 in cell lysates or serum; however, assay conditions must be established by the end user.
