- RNASE2, also known as eosinophil-derived neurotoxin (EDN), RNase UpI-2, ribonuclease 2, and ribonuclease US, is a non-secretory ribonuclease belonging to the pancreatic ribonuclease family.
- It functions as a pyrimidine-specific endonuclease with a slight preference for uridine, and exhibits a variety of biological activities including cytotoxicity, helminthotoxicity, and selective chemotaxis for dendritic cells.
- The protein contains a signal peptide and is a glycoprotein with disulfide bonds; it is subject to nitration, which may modulate its activity.
- Subcellular localization includes lysosomes and cytoplasmic granules, consistent with its roles in degradation and immune cell effector functions.
- Tissue expression is found in liver, lung, spleen, leukocytes, and body fluids, indicating broad distribution and potential roles in both tissue remodeling and host defense.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fragment covering amino acids 28–161, which corresponds to the mature protein after signal peptide cleavage. This design may enhance specificity for the native, processed form.
- For Western blot (WB), a band at approximately 18 kDa is expected; consider additional bands from glycosylation or dimerization under non-reducing conditions.
- In immunofluorescence on paraffin-embedded sections (IF-P), antigen retrieval may be necessary; validate in known expressing tissues such as liver or leukocyte-rich samples.
- For ELISA, the antibody can be considered as a capture or detection reagent; pairing with another clone or antibody may be required for sandwich formats.
