Biological Background: Olig2 Function and Localization
- Oligodendrocyte transcription factor 2 (Olig2), also known as BHLHB1, BHLHE19, PRKCBP2, and RACK17, is a basic helix-loop-helix transcription factor encoded by the OLIG2 gene.
- Essential for oligodendrocyte and motor neuron specification in the spinal cord and hindbrain development.
- Cooperates with ZNF488 to drive oligodendrocyte differentiation and with OLIG1 to establish the pMN domain of the neural tube.
- Functions as a key regulator of progenitor cell fates during cortical gliogenesis and gliomagenesis, including activation of oligodendrocyte precursor cells (OPCs) and repression of GSX2-dependent olfactory bulb interneuron progenitors.
- Antagonizes V2 interneuron development and NKX2-2-induced V3 interneuron differentiation.
- Subcellular localization includes the nucleus and cytoplasm, consistent with its role as a DNA-binding transcription factor.
- Highly expressed in oligodendrocytes and overexpressed in oligodendrogliomas; expression variable in glioblastomas; also linked to chromosomal rearrangements in certain cancers.
- Keywords: chromosomal rearrangement, proto-oncogene, developmental protein, transcription regulation, DNA-binding, nucleus, cytoplasm, Ubl conjugation.
Experimental Guidance and Technical Tips
- This rabbit monoclonal antibody was raised against a synthetic peptide corresponding to human Olig2 and is recommended for Western blot (WB), immunofluorescence (IF-P), immunohistochemistry (IHC-P), and ELISA.
- Given the nuclear and cytoplasmic localization of Olig2, ensure thorough sample lysis and consider nuclear fractionation for stronger signal in WB.
- For immunohistochemistry, test antigen retrieval methods to optimize detection in formalin-fixed paraffin-embedded tissues.
- Validate the antibody in the relevant species (human, mouse, rat) using appropriate positive and negative controls, such as oligodendrocyte-rich brain tissues or cell lines.
- Since Olig2 is a transcription factor, its expression levels may be moderate; dilute the antibody according to the recommended starting range and adjust based on observed signal-to-noise ratio.
