Biological Background: MMP1 Function and Localization
- Interstitial collagenase (MMP-1), also known as fibroblast collagenase, is a member of the matrix metalloproteinase (MMP) family that degrades extracellular matrix components.
- Cleaves fibrillar collagens of types I, II, and III at a single site in the helical domain, as well as non-fibrillar collagens types VII and X, playing a key role in tissue remodeling. Related references: PMID:1645757, PMID:2153297, PMID:2557822
- In the context of HIV infection, MMP1 interacts with and cleaves the secreted viral Tat protein, which can decrease Tat-mediated neurotoxicity. Related references: PMID:16807369
- Localized to the extracellular space and extracellular matrix after secretion, where it exerts its proteolytic activity.
- Contains a signal peptide, an autoinhibitory propeptide with a cysteine switch, a catalytic domain with a zinc-binding motif, and hemopexin-like repeats; activated by autocatalytic cleavage.
- Requires calcium and zinc as cofactors for structural stability and catalytic activity, respectively.
- Annotated with keywords including collagen degradation, host-virus interaction, zymogen, phosphoprotein, and glycoprotein, reflecting its multifaceted regulation and function.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 93–210 of human MMP1, which covers the C-terminal part of the propeptide and the N-terminal region of the catalytic domain; the antibody may recognize both pro and active forms.
- In western blotting, a band around 54 kDa is expected for the full-length proenzyme; additional lower molecular weight bands corresponding to activated forms may appear depending on sample processing and autocatalysis.
- For ELISA, note that MMP1 can be present in latent or active forms; consider assay format and sample handling conditions to ensure optimal detection.
- The antibody cross-reacts with mouse and rat MMP1, enabling its use in rodent models; validate reactivity in your specific tissue or cell line.
- This reagent is suitable for research applications and IVD raw material use; optimize working dilutions for your specific assay system.
