Biological Background: IDH1 Function and Localization

  • Isocitrate dehydrogenase [NADP] cytoplasmic (IDH1), also known as cytosolic NADP-isocitrate dehydrogenase, IDPc, NADP(+)-specific ICDH, and oxalosuccinate decarboxylase.
  • Catalyzes the NADP(+)-dependent oxidative decarboxylation of isocitrate (D-threo-isocitrate) to 2-ketoglutarate, which is required by other enzymes such as phytanoyl-CoA dioxygenase. Plays a critical role in the generation of NADPH, an important cofactor in many biosynthesis pathways. Related references: PMID:10521434 PMID:19935646
  • Subcellular localization includes cytoplasm, cytosol, and peroxisome.
  • May act as a corneal epithelial crystallin and may be involved in maintaining corneal epithelial transparency.
  • Post-translational modifications include acetylation and phosphorylation; the protein binds magnesium or manganese as cofactors and uses NADP.
  • Part of the tricarboxylic acid cycle and glyoxylate bypass; encoded by the IDH1 gene (also known as PICD).

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 100-200 of human IDH1. This region is conserved across human, mouse, and rat, consistent with the observed cross-reactivity.
  • For Western blotting (WB), the expected molecular weight is approximately 47 kDa. Validate detection in relevant cell or tissue lysates from human, mouse, or rat.
  • In immunofluorescence/immunocytochemistry (IF/ICC), expect cytoplasmic and peroxisomal staining patterns. Use appropriate fixation and permeabilization conditions for optimal signal.
  • For ELISA applications, the antibody can be used as a capture or detection reagent; pair with another antibody recognizing a distinct epitope for sandwich assays.
  • Specificity can be confirmed by pre-adsorption with the immunizing peptide or by using IDH1 knockout cell lines.
