Biological Background: hnRNP A1 Function and Localization

  • Heterogeneous nuclear ribonucleoprotein A1 (hnRNP A1), also known as UP1, ALS19, ALS20, or HNRPA1, is a multifunctional RNA-binding protein encoded by the HNRNPA1 gene in humans.
  • Core role in mRNA metabolism: packages pre-mRNA into hnRNP particles, mediates nucleocytoplasmic transport of poly(A) mRNA, and modulates splice site selection, thereby influencing alternative splicing decisions. Related references: PMID:17371836
  • Regulates alternative splicing of pyruvate kinase PKM by binding repressively to sequences flanking PKM exon 9, inhibiting exon 9 inclusion and promoting PKM M2 isoform production. Related references: PMID:20010808
  • Contributes to translational control by binding to the internal ribosome entry site (IRES) of APAF1 mRNA, thereby inhibiting translation of the apoptosis protease activating factor APAF1. Related references: PMID:31498791
  • May interact with specific miRNA hairpins, expanding its regulatory potential in post-transcriptional gene silencing. Related references: PMID:28431233
  • Shuttles between nucleus and cytoplasm; predominantly nuclear but localized to the cytoplasm under certain conditions, consistent with its roles in RNA processing and transport.
  • Subject to multiple post-translational modifications including acetylation, methylation, phosphorylation, and ubiquitin-like conjugation, as suggested by proteomic studies.
  • Implicated in neurodegenerative diseases: mutations in HNRNPA1 are associated with amyotrophic lateral sclerosis (ALS) and inclusion body myopathy with early-onset Paget disease and frontotemporal dementia (IBMPFD).

Experimental Guidance and Technical Tips

  • The antibody was raised against a recombinant protein covering full-length human hnRNP A1 (amino acids 1–372). This broad immunogen may enable recognition of multiple isoforms or splice variants; performance should be validated in the relevant experimental context.
  • For Western blotting, a band corresponding to the predicted molecular weight of ~39 kDa is expected. Consider using whole cell lysates from human, mouse, or rat tissues/cells, as cross-reactivity is confirmed in these species.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimise antigen retrieval and antibody dilution for the specific tissue or cell type, as localisation may be predominantly nuclear or cytoplasmic depending on cellular state.
  • When performing ELISA, ensure that the coating antigen maintains the native conformation or key epitopes present in the immunogen to maximise detection sensitivity.
