Biological Background: HABP4 Function and Localization
- HABP4, also known as intracellular hyaluronan-binding protein 4 or Ki-1/57 intracellular antigen, is a ribosome-binding protein that promotes ribosome hibernation, stabilizing inactive ribosomes and protecting them from proteasomal degradation (By similarity).
- It binds RNA, regulating transcription and pre-mRNA splicing, and specifically binds to poly(U) RNA via its C-terminus. Related references: PMID:14699138, PMID:16455055, PMID:19523114
- HABP4 appears to play a role in the formation of PML nuclear bodies. Related references: PMID:28695742
- It negatively regulates the DNA-binding activity of the transcription factor MEF2C in myocardial cells in response to mechanical stress (By similarity).
- Subcellularly, HABP4 localizes to the nucleus, cytoplasm, stress granules, sarcoplasm, and various nuclear bodies including nucleolus, speckles, Cajal bodies, and gems.
- Tissue expression is highest in brain, heart, and kidney, with moderate expression in skeletal muscle; it is also found in tumor cell lines and activated leukocytes.
- Post-translational modifications include phosphorylation and methylation; it also undergoes ubiquitination (isopeptide bond and Ubl conjugation).
- Alternative splicing generates multiple transcript variants.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide encompassing residues 50–150; thus reactivity is expected against denatured protein, making it suitable for Western blot under reducing conditions.
- For WB, the predicted molecular weight is ~46 kDa; confirm specificity by including appropriate positive (e.g., HeLa, HEK293) and negative controls.
- Cross-reactivity with mouse and rat is reported; validate in the target species’ tissue or cell lysate to ensure equivalent performance.
- For ELISA applications, optimize coating buffer, antibody titer, and detection system empirically to achieve the required dynamic range.
- As a polyclonal antibody, lot-to-lot consistency should be verified for reproducibility in quantitative assays.
- Upon receipt, store the antibody according to the manufacturer’s guidelines; the product is shipped on ice to preserve stability.
