Biological Background: ESE1/ELF3 Function and Localization
- ESE1/ELF3 is an ETS-related transcription factor that binds to the consensus core sequence GGA[AT] and acts as a transcriptional activator.
- It synergizes with POU2F3 to transactivate the SPRR2A promoter and with RUNX1 to transactivate ANGPT1, and also regulates a variety of target genes including CCL20, CLND7, FLG, KRT8, NOS2, PTGS2, TGFBR2, and TGM3.
- Plays a role in epithelial cell differentiation and tumorigenesis, and is a critical downstream effector of the ERBB2 signaling pathway.
- Involved in mediating vascular inflammation and may contribute to inflammatory responses.
- Localizes to both the cytoplasm and nucleus, suggesting dynamic regulation of its transcriptional activity.
- Expressed exclusively in tissues with high content of terminally differentiated epithelial cells, including mammary gland, colon, trachea, kidney, prostate, uterus, stomach, and skin.
- Alternatively spliced isoforms may exist, contributing to functional diversity.
- Contains key structural domains that facilitate DNA binding and transcriptional regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-280 of human ESE1/ELF3; the antibody may recognize the N-terminal region. Validate the epitope if precise mapping is required.
- For Western blot, a band at approximately 41 kDa is expected. Use positive controls such as epithelial cell lines (e.g., MCF-7, HeLa) expressing ESE1/ELF3.
- For immunohistochemistry (paraffin-embedded sections), consider antigen retrieval techniques and validate in tissues with high expression levels (e.g., colon, mammary gland).
- As a polyclonal antibody, batch-to-batch consistency should be verified in the intended assay system. Including appropriate positive and negative controls is recommended.
