Biological Background: DNA Polymerase gamma Function and Localization
- DNA polymerase subunit gamma-1 (POLG) is the catalytic core of mitochondrial DNA polymerase, responsible for the replication of the entire mitochondrial genome. It synthesizes both heavy and light strands using single-stranded DNA templates and RNA primers, functioning as a highly processive replisome together with TWNK helicase and SSBP1. - POLG possesses a 3'→5' exonuclease proofreading activity that corrects base substitutions, single-base additions, and deletions, ensuring high fidelity during mtDNA replication. - After completing mtDNA synthesis, POLG mediates 3'→5' exonucleolytic degradation at the nick to facilitate proper ligation, and it also exhibits 5'-deoxyribose phosphate (dRP) lyase activity, enabling its participation in the mitochondrial base-excision repair pathway. - Mutations in the POLG gene are linked to a spectrum of mitochondrial disorders, including progressive external ophthalmoplegia (PEO), neuropathies, epilepsy, and Leigh syndrome. It is a primary mitochondrial disease gene associated with mitochondrial DNA depletion and deletion syndromes.
- The protein localizes to the mitochondrial matrix and is a component of the mitochondrial nucleoid, where it associates with mtDNA to facilitate replication and maintenance.
- POLG is a large, multi-functional enzyme (approximately 140 kDa) that belongs to the DNA polymerase type-A family. It requires magnesium ions for polymerase and exonuclease activities.
Experimental Guidance and Technical Tips
- The immunogen used to raise this antibody corresponds to a peptide within the C-terminal region (amino acids 1140–1239) of human POLG. This epitope lies outside the catalytic domains and may provide recognition of both wild-type and some mutant forms. However, for each experimental system, verify epitope accessibility under native and denaturing conditions.
- For Western blot, the predicted molecular weight is 140 kDa. Use appropriate gel percentage and transfer conditions for high molecular weight proteins, and include a positive control such as mitochondrial lysates from human or mouse cells.
- Cross-reactivity with mouse POLG has been confirmed, making this antibody suitable for studies in mouse models of mitochondrial disease. Due to high sequence conservation, cross-reactivity with other species may be possible but should be empirically validated.
- For ELISA, the antibody can be used as a capture or detection reagent. Titrate the antibody to determine the optimal working concentration, and ensure the coating antigen contains the C-terminal epitope or a recombinant POLG fragment spanning the immunogen region.
- Note that POLG expression is ubiquitous but enriched in tissues with high energy demand. Consider validating detection sensitivity in relevant sample systems, such as liver, muscle, or brain mitochondrial preparations.
