Biological Background: DNA Ligase IV Function and Localization
- DNA ligase 4 (LIG4), also known as DNA ligase IV or polydeoxyribonucleotide synthase [ATP] 4, is encoded by the LIG4 gene and is a member of the ATP-dependent DNA ligase family.
- LIG4 catalyzes the final ligation step in non-homologous end joining (NHEJ), an essential pathway for repairing DNA double-strand breaks (DSBs) and for V(D)J recombination during immune development. Related references: PMID:12517771, PMID:17290226, PMID:23523427, PMID:9242410
- LIG4 forms a tight subcomplex with the scaffold protein XRCC4, which markedly stimulates its ligation activity; the LIG4-XRCC4 complex can seal single-strand nicks and, with lower efficiency, join ends bearing 2-nucleotide microhomology or 1-nucleotide gaps. Related references: PMID:17290226, PMID:9242410, PMID:9809069
- Efficient loading of the LIG4-XRCC4 complex onto DNA breaks requires the assembly of the DNA-dependent protein kinase (DNA-PK) holoenzyme at the damage site. Related references: PMID:10854421
- LIG4 also regulates the nuclear accumulation of XRCC4, underscoring a dynamic interplay that localizes the NHEJ machinery to sites of DNA damage. Related references: PMID:24984242
- The protein is predominantly nuclear, consistent with its function in genomic DNA repair, and is expressed at highest levels in testis, thymus, prostate, and heart.
- With a predicted molecular mass of ~104 kDa, LIG4 contains conserved motifs for ATP binding, magnesium/metal coordination, and tandem BRCT repeats that mediate protein–protein interactions in DNA repair complexes.
Experimental Guidance and Technical Tips
- The immunogen used for this antibody is a recombinant fragment spanning amino acids 1–210 of human LIG4 (NP_002303.2); when designing experiments, consider that this N‑terminal region may be conserved across species, supporting the observed cross‑reactivity.
- Validated applications include western blotting (WB), immunohistochemistry on paraffin‑embedded sections (IHC-P), and ELISA; for WB, the predicted molecular weight is approximately 104 kDa—always validate detection in the relevant cell or tissue lysate.
- Cross‑reactivity is reported for human, mouse, and rat; for experiments in other species, evaluate sequence homology of the immunogen region and include appropriate controls to confirm specificity.
