Anti-Dengue virus (NS1)
Target Introduction:
Dengue virus (DENV) is a mosquito-borne virus belonging to the Flaviviridae family. Its RNA genome encodes three structural and seven nonstructural (NS) proteins. Nonstructural protein 1 (NS1) is a multifunctional protein actively secreted by both vertebrate and mosquito cells during infection. In mosquito cells, NS1 secretion occurs unconventionally, dependent on caveolin-1.
Secretion Mechanism in Mosquito Cells:
NS1 associates with and relies on the caveolin chaperone complex (CCC) for secretion in mosquito cells. The CCC comprises caveolin-1 and the chaperones FKBP52, Cy40, and CyA, responsible for cholesterol trafficking inside the cell. Classic secretion inhibitors (e.g., brefeldin A, Golgicide A, and Fli-06) don't significantly impact NS1 secretion but significantly reduce recombinant luciferase secretion and virion release in mosquito cells. Silencing CAV-1 or FKBP52, or inhibiting CyA, decreases NS1 secretion without affecting virion release. NS1 colocalizes and interacts with all CCC proteins, and CAV-1 and FKBP52 expression is augmented in DENV-infected cells. Zika virus (ZIKV) NS1 follows the same secretory pathway as DENV NS1 in mosquito cells.
