Biological Background: CYP1B1 Function and Localization
- Cytochrome P450 1B1 (CYP1B1) is a monooxygenase that inserts one oxygen atom into substrates using NADPH via cytochrome P450 reductase, metabolizing fatty acids, steroid hormones (estradiol, testosterone, progesterone), vitamins, and xenobiotics including procarcinogens (PubMed:10681376, PubMed:11555828, PubMed:12865317, PubMed:10426814). Related references: PMID:10681376 PMID:11555828 PMID:12865317 PMID:10426814
- Also known as CYPIB1, CP1B, ASGD6, GLC3A, and P4501B1.
- Catalyzes the 4-hydroxylation of 17beta-estradiol, epoxidizes polyunsaturated fatty acids to epoxyeicosatrienoic acids (EpETrE), and exhibits dehydratase activity toward hydroperoxyeicosatetraenoates (HpETEs) independent of NADPH and O2 (PubMed:11555828, PubMed:12865317, PubMed:20972997, PubMed:21068195). Related references: PMID:11555828 PMID:12865317 PMID:20972997 PMID:21068195
- Key for all-trans retinoic acid biosynthesis in extrahepatic tissues via two-step oxidation of all-trans retinol (PubMed:10681376, PubMed:15258110). Related references: PMID:10681376 PMID:15258110
- Subcellularly localized to the endoplasmic reticulum membrane, microsome membrane, and mitochondrion.
- Expressed in multiple tissues including heart, brain, lung, kidney, testis, ovary, and retinal endothelial cells (PubMed:8175734, PubMed:19005183). Related references: PMID:8175734 PMID:19005183
- Associated with glaucoma and Peters anomaly according to UniProt keywords.
- Contains a heme iron cofactor and an experimentally determined 3D structure.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal fragment (aa 244–543), so the antibody may recognize both native and denatured CYP1B1. Validate in relevant sample systems expressing human CYP1B1.
- In Western Blot, anticipate a band near 61 kDa; confirm specificity using positive controls (e.g., CYP1B1-overexpressing lysates) and negative controls.
- For IF/ICC, expect staining consistent with ER/mitochondrial localization; consider co-staining with organelle markers to verify distribution.
- When using ELISA, optimize coating conditions and detection format; the polyclonal nature can provide signal amplification but may require careful blocking to reduce background.
- Cross-reactivity is reported for human CYP1B1; for other species, verify sequence homology of the immunogen region if cross-species detection is desired.
