Biological Background: BRD9 Function and Localization
- BRD9 (Bromodomain-containing protein 9), also known as Rhabdomyosarcoma antigen MU-RMS-40.8, plays a role in chromatin remodeling and regulation of transcription. Related references: PMID:22464331, PMID:26365797
- It acts as a chromatin reader that specifically recognizes and binds acylated histones, including those that are acetylated and butyrylated. Related references: PMID:26365797
- BRD9 is a core component of the GBAF (GLTSCR1/1L-containing BAF) subcomplex of the SWI/SNF chromatin remodeling complex, which uses ATP hydrolysis to modulate DNA-histone contacts within nucleosomes. Related references: PMID:29374058

Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning the C-terminal region (aa 274–597) of human BRD9, which encompasses the bromodomain. Given the high conservation of this domain, cross-reactivity with mouse and rat is observed.
- For Western blotting, a band near 67 kDa is expected. Using nuclear extracts from human, mouse, or rat cell lines may enhance detection due to the nuclear localization of BRD9.
- In ELISA applications, the antibody can be considered as a capture or detection reagent; optimization with recombinant BRD9 protein or cell lysates is advised.
- Always validate the antibody in the relevant sample system and perform appropriate controls to confirm specificity.
