25-hydroxy vitamin D (Anti-Vitamin 25 OH)
Target Introduction:
25-hydroxy vitamin D (25 OH Vitamin D), also known as calcidiol, is a prehormone produced in the liver by hydroxylation of vitamin D (either vitamin D2 or vitamin D3). Measuring the level of 25-hydroxy vitamin D in the blood is the best way to determine a person's vitamin D status. This assay utilizes a monoclonal antibody that recognizes both 25 OH Vitamin D2 and 25 OH Vitamin D3. The normal range is generally considered to be between 20-40 ng/mL (60-100 nmol/L) or 30-50 ng/mL (75-125 nmol/L), although specific ranges may vary between laboratories.
- 25-hydroxy vitamin D3 (cholecalciferol): Derived from the body's own production, animal sources, or cholecalciferol supplements.
- 25-hydroxy vitamin D2 (ergocalciferol): Derived from fortified foods or ergocalciferol supplements. The total 25 hydroxy vitamin D level in your blood is important.
Application:
- ELISA: The described monoclonal antibody (Anti-Vitamin 25 OH D2/D3) is specifically designed for use in ELISA assays.
- Dry Immunofluorescence Assay: Can be used with Getein1100 analyzer
Relationship with Related Diseases:
Abnormal 25-hydroxy vitamin D levels can indicate a vitamin D deficiency or excess, which can be associated with various health conditions. Lower-than-normal levels can be due to:
- Lack of sun exposure, darkly pigmented skin, or sunscreen use
- Insufficient vitamin D in the diet
- Liver and kidney diseases
- Poor food absorption
- Use of certain medications (e.g., phenytoin, phenobarbital, rifampin)
- Poor vitamin D absorption due to advanced age, weight-loss surgery, or conditions affecting fat absorption.
Antibody Specificity and Cross-Reactivity:
- The antibody recognizes both natural 25 OH Vitamin D2 and 25 OH Vitamin D3.
- Cross-reactivity:
- Vitamin D3 < 3 %
- Vitamin D2 ~ 1 %
- 1, 25(OH)2 Vitamin D3* ~ 20 % (*low concentration in blood, about 0.1 %)
- 1, 25(OH)2 Vitamin D2* ~ 2 %
