Biological Background: YY1 Function and Localization
- YY1 (Yin Yang 1), also known as Delta transcription factor, NF-E1, INO80S, and UCRBP, is a multifunctional transcription factor that exhibits both repressor and activator functions depending on the promoter context and interacting cofactors.
- It binds to the consensus DNA sequence 5'-CCGCCATNTT-3' near transcription start sites; methylation of the initial CG dinucleotide greatly reduces binding affinity. Related references: PMID:15329343 PMID:17721549 PMID:24326773
- In the absence of adenovirus E1A protein, YY1 acts as a repressor, but in its presence, it switches to an activator, demonstrating its context-dependent transcriptional regulation. Related references: PMID:1655281
- YY1 synergizes with SMAD1 and SMAD4 in bone morphogenetic protein (BMP)-mediated cardiac-specific gene expression by binding to SMAD binding elements within BMP response elements. Related references: PMID:15329343
- It recruits the PRC2/EED-EZH2 complex to mediate transcriptional repression and also interacts with the PR-DUB complex, targeting specific gene-regulatory regions for deubiquitination. Related references: PMID:11158321 PMID:20805357
- YY1 is proposed as a core component of the chromatin remodeling INO80 complex and is involved in DNA repair, in vitro binding to Holliday junction recombination intermediates. Related references: PMID:18026119 PMID:28575647
- Subcellularly, YY1 localizes to the nucleus matrix, consistent with its role in transcriptional regulation and chromatin organization.
- Post-translational modifications of YY1 include phosphorylation, ADP-ribosylation, and ubiquitination; the protein contains multiple zinc-finger domains essential for DNA binding.
Experimental Guidance and Technical Tips
- The immunogen targeting amino acids 80–160 of human YY1 may detect both full-length and truncated isoforms; validate the observed band pattern in your specific sample system.
- For Western blotting, the predicted molecular weight is ~45 kDa; optimize blocking conditions and antibody dilution using whole-cell lysates from human, mouse, or rat cells.
- In ChIP assays, include appropriate positive and negative control loci (e.g., known YY1 target promoters vs. intergenic regions) and confirm enrichment with qPCR.
- When using ELISA, consider cross-reactivity testing against other zinc-finger transcription factors if non-specific signals are observed.
