Biological Background: YOD1 Function and Localization
- YOD1 (Ubiquitin thioesterase OTU1, also known as DUBA8, OTUD2) is a member of the OTU family of deubiquitinases.
- It functions as a hydrolase that removes conjugated ubiquitin from proteins, primarily acting on 'Lys-27'-, 'Lys-29'-, and 'Lys-33'-linked polyubiquitin chains, and also hydrolyzes 'Lys-11'-linked chains.
- Key role in endoplasmic reticulum-associated degradation (ERAD): it trims the ubiquitin chain on misfolded lumenal substrates to facilitate their threading through the VCP/p97 pore.
- Localizes to the cytoplasm.
- May participate in macroautophagy, including the clearance of damaged lysosomes. It may recruit PLAA, UBXN6 and VCP to damaged lysosome membranes decorated with K48-linked ubiquitin chains and remove these chains allowing autophagosome formation. Related references: PMID:27753622
- Contains a OTU domain and a zinc-finger domain, classified as a thiol protease requiring metal binding.
