Biological Background: YES1 Function and Localization
- YES1 encodes a non-receptor tyrosine kinase of the Src family, functioning as a proto-oncogene (c-Yes) and playing critical roles in cell growth, survival, apoptosis, cell-cell adhesion, cytoskeleton remodeling, and differentiation.
- Upon stimulation by receptor tyrosine kinases (e.g., EGFR, PDGFR, CSF1R, FGFR), YES1 is recruited to phosphorylated receptors and phosphorylates downstream substrates such as PARD3 (promoting epithelial tight junction assembly) and CDK4 (regulating G1 phase cell cycle progression).
- YES1 participates in T-cell migration by phosphorylating DPYSL2 downstream of CXCL12 stimulation, and acts in the CD95L/FASLG signaling pathway to mediate AKT-dependent cell migration.
- It catalyzes phosphorylation of the organic cation transporter OCT2, thereby enhancing its transport activity. Related references: PMID:26979622
- The protein localizes to the cell membrane, cytoplasm, cytoskeleton, centrosome, and cell junctions, reflecting its diverse signaling roles.
