Biological Background: XBP1 Function and Localization
- XBP1 (X-box-binding protein 1), also known as TREB5 or XBP2, is a transcription factor that exists as two isoforms generated by unconventional cytoplasmic splicing. Isoform 1 is the unspliced form and acts as a weak transcriptional factor, while isoform 2 is the spliced, potent transcriptional activator. Related references: PMID:19394296, PMID:21233347, PMID:23529610, PMID:16461360, PMID:25239945, PMID:8657566
- Isoform 2 functions as a master regulator of the unfolded protein response (UPR) during ER stress. It binds to UPRE and ERSE elements to induce genes encoding ER chaperones and ER-associated degradation (ERAD) components, thereby enhancing the protein folding capacity and maintaining ER homeostasis. Related references: PMID:11779464, PMID:25239945, PMID:15466483, PMID:23529610, PMID:19416856, PMID:23184933, PMID:25190803, PMID:25448679
- Isoform 1 acts as a negative feedback regulator by recruiting its own mRNA for splicing and promoting proteasome-mediated degradation of isoform 2, thus preventing constitutive UPR activation. Related references: PMID:16461360, PMID:25239945
- Plays a critical role in terminal differentiation of B lymphocytes to plasma cells and in immunoglobulin production, with expression observed in plasma cells within rheumatoid synovium. Related references: PMID:11460154
- In endothelial cells, XBP1 contributes to VEGF-induced proliferation and angiogenesis, modulates disturbed flow responses via HDAC3, and regulates autophagy and apoptosis. Related references: PMID:23529610, PMID:25190803, PMID:19416856, PMID:23184933
- Localizes to the endoplasmic reticulum (membrane), nucleus, and cytoplasm, with isoform 2 predominantly nuclear upon ER stress.
- Tissue expression includes plasma cells, proliferating endothelial cells, and is overexpressed in primary and metastatic breast cancer cells. Related references: PMID:11460154, PMID:25280941, PMID:19416856
- Post-translational modifications include acetylation, phosphorylation, cleavage, and ubiquitination; alternative splicing generates functionally distinct isoforms.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein; consider validating linear vs. conformational epitope recognition in your specific assay.
- For Western blot, the expected molecular weight is 29 kDa for the unspliced isoform (XBP1u). Detection of the spliced isoform (XBP1s) may require optimized gel conditions and validation in your experimental system.
- The antibody is suitable for immunoprecipitation and ELISA. For IP, test both native and cross-linked conditions depending on target complex interactions.
- Given cross-reactivity with human and mouse, include positive and negative controls from both species to verify specificity in your sample type.
