  • VPS37A, also known as Hepatocellular carcinoma-related protein 1 (HCRP1) or ESCRT-I complex subunit VPS37A, is a component of the ESCRT-I complex, a key regulator of vesicular trafficking.
  • It is required for the sorting of endocytic ubiquitinated cargos into multivesicular bodies (MVBs), a critical step in the endosomal-lysosomal degradation pathway.
  • The protein may play a role in cell growth and differentiation, as suggested by its functional characterization.
  • VPS37A localizes to the late endosome membrane and also to the nucleus, indicating potential shuttling or dual functions.
  • It is widely expressed in tissues, including heart, brain, placenta, liver, skeletal muscle, kidney, and pancreas, with particularly high abundance in liver. Notably, its expression is strongly decreased or undetected in hepatomas, linking it to hepatocellular carcinoma.
  • The gene is associated with hereditary spastic paraplegia (SPG53) and undergoes alternative splicing. The protein is a phosphoprotein and has been identified in proteomics studies.
