Biological Background: VISTA Function and Localization
- VISTA (V-type immunoglobulin domain-containing suppressor of T-cell activation) functions as an immunoregulatory receptor that inhibits T-cell responses. Related references: PMID:24691993
- Alternative names include platelet receptor Gi24, stress-induced secreted protein-1 (Sisp-1), and V-set domain-containing immunoregulatory receptor.
- It may promote embryonic stem cell differentiation by inhibiting BMP4 signaling (by similarity).
- VISTA may stimulate MMP14-mediated activation of MMP2. Related references: PMID:20666777
- The protein localizes to the cell membrane as a single-pass transmembrane receptor.
- It is expressed in spleen, on myeloid cells (e.g., CD11b+ monocytes, CD66b+ neutrophils), at low levels on CD4+ and CD8+ T-cells and a subset of NK cells, but not on B cells. Higher expression is seen in placenta, lung, and plasma blood leukocytes.
- VISTA is a glycoprotein and phosphoprotein, containing an immunoglobulin domain and disulfide bonds.
Experimental Guidance and Technical Tips
- For western blotting, consider testing human spleen or lung lysates where VISTA expression is relatively high; validate the band at the expected molecular weight (~34 kDa).
- The immunogen covers the extracellular domain (amino acids 33–194), making the antibody potentially suitable for ELISA detection of soluble or cell-surface VISTA; optimize coating and detection conditions in the relevant sample matrix.
- Due to the polyclonal nature, it may be advisable to include appropriate controls (e.g., lysates from VISTA-overexpressing cells vs. knockdown/knockout) to confirm specificity.
- Cross-reactivity with other species is not indicated; verify reactivity if used with non-human samples.
