Biological Background: VIP Function and Localization
- VIP (Vasoactive Intestinal Peptide) is a neuropeptide encoded by the VIP gene, belonging to the glucagon/secretin superfamily. The preproprotein is proteolytically processed to yield multiple bioactive peptides, including VIP and PHM-27 (peptide histidine methionine 27).
- Through binding to its cognate G protein-coupled receptors VPAC1 (VPR1) and VPAC2 (VPR2), VIP mediates pleiotropic effects including vasodilation, smooth muscle relaxation, stimulation of myocardial contractility, and regulation of glycogenolysis. - The immunogen used is a recombinant fragment spanning amino acids 31-170 of human VIP, which covers the mature peptide region. For Western blot, a band around 19 kDa is expected, but additional bands corresponding to precursor or degradation products may also be observed.
- For immunohistochemistry (IHC-P), antigen retrieval methods may be necessary to unmask the epitope in formalin-fixed paraffin-embedded tissues. Validate staining specificity using positive control tissues such as brain, colon, or lung where VIP is known to be expressed.
- In ELISA applications, consider using a competitive or sandwich format with appropriate standards to quantify VIP levels in serum or tissue extracts.
- As a polyclonal antibody, lot-to-lot variability may occur; it is advisable to titrate the antibody in your specific assay conditions for optimal performance.
