Biological Background: USP9X Function and Localization

  • USP9X (Ubiquitin carboxyl-terminal hydrolase 9X), also known as FAF, FAM, hFAM, DFFRX, MRX99, is a large deubiquitinase enzyme encoded by the USP9X gene located on the X chromosome. It belongs to the peptidase C19 family of ubiquitin-specific proteases.
  • Acting as a thiol protease, USP9X specifically hydrolyzes polyubiquitin chains with a preference for Lys-11 linkages, followed by Lys-63, Lys-48, and Lys-6 linkages, thereby removing ubiquitin from target proteins and regulating protein turnover. Related references: PMID:30914461
