Biological Background: USP7/HAUSP Function and Localization

  • USP7, also known as HAUSP, is a deubiquitinating enzyme that removes ubiquitin from target proteins, regulating their stability and activity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a large central region (amino acids 206-1102), which may detect full-length USP7 and potentially some isoforms. Confirm specificity by Western blot using appropriate lysates (human, mouse, rat) and consider siRNA knockdown controls.
  • For Western blot, a band around 128 kDa is expected. Use reducing conditions and standard transfer protocols; note that USP7 may undergo post-translational modifications that slightly alter migration.
  • For immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), optimize fixation and permeabilization conditions. Antigen retrieval may be required for IHC-P. Validate with positive and negative tissue/cell controls.
  • Since this is a polyclonal antibody, lot-to-lot variation may occur; consider validating each new lot for consistency in target applications.
