Biological Background: USP7/HAUSP Function and Localization

USP7/HAUSP is a deubiquitinating enzyme that removes ubiquitin moieties from target proteins, regulating their stability and function. It preferentially cleaves 'Lys-48'-linked ubiquitin chains (PubMed:22689415).

USP7 plays a dual role in the p53 pathway: it deubiquitinates and stabilizes p53, enhancing p53-mediated transcription and apoptosis (PubMed:11923872, PubMed:25283148, PubMed:26786098), and also deubiquitinates MDM2, preventing its self-ubiquitination and promoting MDM2-mediated p53 degradation (PubMed:15053880, PubMed:16845383, PubMed:18566590).
