Biological Background: USP10 Function and Localization

  • USP10 (Ubiquitin carboxyl-terminal hydrolase 10) is a thiol protease that functions as a deubiquitinating enzyme, removing ubiquitin from various substrates to regulate protein stability, signaling, and trafficking. Related references: PMID:11439350 PMID:18632802 PMID:31981475

  • In unstressed cells, USP10 localizes to the cytoplasm where it specifically deubiquitinates and stabilizes p53/TP53 by counteracting MDM2-mediated degradation. Upon DNA damage, USP10 translocates to the nucleus to further deubiquitinate p53 and facilitate the DNA damage response. Related references: PMID:20096447

  • USP10 controls autophagy by deubiquitinating BECN1, a key autophagy regulator, thereby stabilizing PIK3C3/VPS34 complexes. This creates a regulatory loop where PIK3C3/VPS34 complexes, in turn, regulate USP10 stability, impacting p53 levels. Related references: PMID:21962518

  • USP10 plays a critical role in ribosome quality control and stress granule dynamics. It inhibits stress granule assembly by lowering G3BP1/2 valency and mediates deubiquitination of 40S ribosomal proteins (RPS2, RPS3, RPS10) to promote subunit recycling after translation stalling. Related references: PMID:27022092 PMID:32302570 PMID:31981475 PMID:34348161 PMID:34469731

  • USP10 deubiquitinates CFTR in early endosomes, enhancing its endocytic recycling and cell surface expression. Related references: PMID:19398555

  • USP10 acts as a negative regulator of NF-κB signaling by deubiquitinating IKBKG or TRAF6 in response to IL-1β or DNA damage, and also blocks RIG-I-mediated MAVS activation during RNA virus infection by removing unanchored K63-linked polyubiquitin from MAVS. Related references: PMID:25861989 PMID:37582970

  • USP10 is widely expressed and localizes to the cytoplasm, nucleus, and early endosomes. Post-translational modifications include acetylation and phosphorylation, and alternative splicing produces multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 499–798) of human USP10. This region may recognize multiple isoforms arising from alternative splicing.
  • For Western blotting, the predicted molecular weight is ~87 kDa; verify band specificity with appropriate controls, especially in samples with active deubiquitinase processes.
  • For immunofluorescence (IF/ICC), note that USP10 shuttles between cytoplasm and nucleus. Consider using cell fractionation or carefully optimized fixation/permeabilization protocols to capture both localizations.
  • The antibody is validated for WB, IF/ICC, and ELISA. Optimize working dilutions empirically for each application and sample type.
  • Because USP10 is involved in stress responses (e.g., DNA damage, ribosome stalling), include appropriate positive and negative controls in your experiments.
