Biological Background: USP10 Function and Localization
- USP10, also known as ubiquitin carboxyl-terminal hydrolase 10, deubiquitinating enzyme 10, or ubiquitin thioesterase 10, is a thiol protease belonging to the ubiquitin-specific processing protease family.
- It functions as a hydrolase that specifically removes conjugated ubiquitin from target proteins, playing a critical role in regulating protein stability and signaling.
- USP10 is a key regulator of p53/TP53: it deubiquitinates and stabilizes p53 in the cytoplasm under unstressed conditions, and translocates to the nucleus upon DNA damage to further regulate p53-dependent DNA damage responses. Related references: PMID:20096447
- It also deubiquitinates BECN1, a key autophagy regulator, thereby stabilizing PIK3C3/VPS34-containing complexes and contributing to a regulatory loop that controls autophagy and p53 levels. Related references: PMID:21962518
- USP10 participates in ribosome quality control by deubiquitinating 40S ribosomal subunit proteins (RPS2/us5, RPS3/us3, RPS10/eS10) to promote subunit recycling after translation stalling. Related references: PMID:31981475, PMID:34348161, PMID:34469731
- Subcellularly, USP10 localizes to the cytoplasm, nucleus, and early endosomes, consistent with its diverse functional roles.
- USP10 undergoes post-translational modifications including acetylation and phosphorylation, and alternative splicing generates multiple isoforms.
- It is widely expressed across tissues and is implicated in processes such as DNA repair, NF-kappa-B signaling attenuation, and negative regulation of RLR antiviral signaling.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human USP10, which may contribute to recognizing the protein across isoforms. Validate specificity using knockdown or knockout controls.
- For Western blot, expect a band at ~87 kDa (predicted molecular weight). Optimize loading amounts and antibody dilution for your sample type.
- For IF/ICC, consider co-staining with markers for cytoplasm, nucleus, and endosomes to reflect USP10's multiple localizations.
- ELISA can be used for quantifying USP10 levels in cell lysates or recombinant protein; include appropriate standards and negative controls.
