Biological Background: Uromodulin Function and Localization

  • Uromodulin (also known as Tamm-Horsfall urinary glycoprotein, THP) is the most abundant protein in normal human urine and is encoded by the UMOD gene.
  • It contributes to the biogenesis and organization of the apical membrane in thick ascending limb of Henle's loop (TALH) epithelial cells, forming a gel-like structure that may influence water barrier permeability.
  • Acts as a receptor for binding and endocytosis of cytokines (IL-1, IL-2) and TNF, linking it to renal immune responses. Related references: PMID:3498215
  • Facilitates neutrophil migration across renal epithelia, potentially modulating inflammatory cell trafficking in the kidney. Related references: PMID:20798515
  • Protects against urinary tract infections by binding to type 1 fimbriated E. coli via the bacterial adhesin fimH, promoting bacterial aggregation and preventing urothelial adhesion, with activity also against K. pneumoniae, P. aeruginosa, and S. mitis. Related references: PMID:11134021 PMID:32616672
  • Localization includes apical and basolateral cell membranes, cilium membrane, and secretion into the urine, consistent with roles both at the cell surface and in the extracellular milieu.
  • Expressed exclusively in kidney epithelial cells of the TALH and distal convoluted tubule, highlighting its specialized renal function.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the central region (aa 175–324) of human Uromodulin, which lies within the ZP domain and is conserved; consider validating reactivity in your species and sample type of interest.
  • For WB, multiple bands may be observed (~55–115 kDa) due to glycosylation and proteolytic processing; include appropriate molecular weight markers and consider using deglycosylation controls if band pattern clarification is needed.
  • This antibody is listed for WB, IF-P, IHC-P, and ELISA; for tissues or cells with low expression, optimize detection conditions and verify with relevant positive and negative controls.
  • Cross-reactivity is reported for human and mouse; if using mouse models, confirm performance in your specific strain and tissue context.
