Biological Background: UPF1/RENT1 Function and Localization
-
UPF1 (Regulator of nonsense transcripts 1), also known as RENT1 or hUpf1, is an ATP-dependent RNA helicase and master regulator of nonsense-mediated mRNA decay (NMD), a surveillance pathway that degrades mRNAs harboring premature termination codons. Related references: PMID:11163187 PMID:16086026 PMID:18172165
-
The enzyme catalyzes ATP-dependent unwinding of RNA:DNA, RNA:RNA, and DNA:DNA duplexes in the 5'-3' direction, with single-stranded DNA strongly stimulating its ATPase activity. Related references: PMID:10999600 PMID:18066079 PMID:21419344 PMID:23275559 PMID:26138914 PMID:30218034
-
Upon translation termination, UPF1 is recruited to stalled ribosomes and cycles between phosphorylated and dephosphorylated states; its phosphorylation by SMG1 is a critical switch that triggers NMD. Related references: PMID:11544179 PMID:19417104 PMID:25220460
-
In the canonical EJC-dependent pathway, UPF1 assembles into a heterotrimeric SURF complex and then with UPF2/UPF3 to form the active surveillance complex, which directs target mRNAs for exonucleolytic and endonucleolytic decay. Related references: PMID:21419344 PMID:12554878
-
UPF1 additionally operates in alternative NMD pathways independently of UPF2/UPF3 and mediates replication-dependent histone mRNA decay at the end of S phase. Related references: PMID:16086026 PMID:18172165 PMID:18447585
-
The protein localizes to the cytoplasm, processing bodies (P-bodies), nucleus, and perinuclear region.
-
UPF1 is ubiquitously expressed, contains zinc-finger domains, and is subject to post-translational modifications including phosphorylation, methylation, and isomerization.
Experimental Guidance and Technical Tips
- As the immunogen corresponds to the C-terminal region (aa 819–1118), the antibody is expected to recognize all UPF1 isoforms and is suitable for detecting full-length and truncated variants in WB and IP.
- For western blot, the predicted molecular weight is ~124 kDa; use reducing conditions and validate with appropriate positive (e.g., HeLa, HEK293) and negative controls.
- In immunofluorescence/ICC, UPF1 typically shows a diffuse cytoplasmic and nuclear punctate pattern; permeabilization conditions may need optimization.
- The reported cross-reactivity with mouse and rat makes this antibody useful for rodent model studies, but validation in the specific sample system is recommended.
- For ELISA and IP applications, the polyclonal nature may provide high sensitivity, though titration and blocking optimization are advisable.
