Biological Background: UNC5C Function and Localization
- UNC5C (Netrin receptor UNC5C), also known as UNC5H3, is a transmembrane receptor belonging to the UNC-5 family.
- It functions as a netrin-dependent axon guidance receptor, mediating repulsion of neuronal growth cones during nervous system development; NTN1/netrin-1 binding may dissociate UNC5C from polymerized TUBB3, increasing microtubule dynamics and axon repulsion. Related references: PMID:28483977
- UNC5C may collaborate with DSCAM in NTN1-mediated axon repulsion independently of DCC, and is involved in corticospinal tract axon guidance and dorsal root ganglion axon projection towards the spinal cord. Related references: PMID:28483977
- Acts as a dependence receptor, inducing apoptosis in the absence of netrin ligand.
- Subcellular localization includes the cell membrane, synapses, growth cones, lamellipodia, and filopodia, consistent with its role in axon guidance.
- Predominantly expressed in the brain, particularly in temporal lobe cortical neurons and hippocampal pyramidal layer. Also detected in kidney but not in developing or adult lung. Related references: PMID:25419706 PMID:9782087
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation, which may regulate its function.
- Associated with neurodegenerative processes such as Alzheimer disease and amyloidosis, as indicated by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 300-360, a region that may influence epitope accessibility; consider using a blocking peptide or competitive assay to confirm specificity.
- For WB, the predicted molecular weight is 103 kDa; however, due to glycosylation and potential cleavage, additional bands may be observed. Validate with positive controls from relevant tissues (e.g., brain lysates from human, mouse, or rat).
- In ELISA, optimize coating concentration and antibody dilution using purified recombinant protein or cell lysates with known UNC5C expression.
- Since UNC5C expression is tissue-specific, inclusion of appropriate negative controls (e.g., lung lysates) is recommended.
- As a polyclonal antibody, lot-to-lot variability is possible; always validate performance in your experimental system.
