Biological Background: UBE2D1 Function and Localization

  • UBE2D1 (Ubiquitin-conjugating enzyme E2 D1), also known as UBCH5, UBC4/5, SFT, or E2(17)KB1, is a member of the ubiquitin-conjugating enzyme family encoded by the UBE2D1 gene.
  • It accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to target proteins, including forming 'Lys-48'-linked polyubiquitin chains. Related references: PMID:22496338, PMID:20061386
  • This enzyme mediates the selective degradation of short-lived and abnormal proteins and participates in the E6/E6-AP-dependent ubiquitination of p53/TP53.
  • UBE2D1 ubiquitinates a range of substrates, including PEX5, STUB1, TRAF6, TRIM63/MURF1, HSP90AB1, and CYP3A4, and undergoes auto-ubiquitination. Related references: PMID:18042044, PMID:18359941, PMID:19103148
  • It lacks inherent specificity for any particular lysine residue of ubiquitin, which allows the generation of diverse ubiquitin chain topologies. Related references: PMID:18042044
  • Essential for the viral activation of IRF3, highlighting a role in innate immunity. Related references: PMID:19854139
  • UBE2D1 is predominantly located in the cytoplasm and is ubiquitously expressed, with elevated levels in the liver of iron-overloaded hemochromatosis patients.

Experimental Guidance and Technical Tips

  • The immunogen (aa 1-147 of human UBE2D1) covers nearly the full-length protein, making the antibody suitable for detecting both native and denatured forms in WB and IHC-P.
  • For WB, the predicted protein weight is 17 kDa; consider using appropriate molecular weight markers and optimize loading controls for small proteins.
  • For IHC-P, validate fixation and antigen retrieval conditions for your specific tissue types; cytoplasmic staining is expected as positive signal.
  • For ELISA, titrate the antibody concentration using the recombinant immunogen as a reference standard.
  • Reported cross-reactivity with mouse and rat should be confirmed by sequence alignment and preliminary experiments in your sample system.
  • Given the ubiquitous expression of UBE2D1, include negative controls (tissue/cell line with low or absent expression) or use knockdown/knockout validation to ensure specificity.
