• TWIST2 (also known as Dermo-1, bHLHa39) is a member of the class A basic helix-loop-helix (bHLH) transcription factor family.
• It functions as a transcriptional repressor by binding to E-box consensus sequences (5'-CANNTG-3'), inhibiting the activity of myogenic factors MYOD1, MYOG, MEF2A, and MEF2C.
• TWIST2 plays a key role in repressing pro-inflammatory cytokines such as TNFA and IL1B.
• It inhibits premature or ectopic differentiation of preosteoblast cells during osteogenesis, possibly modulating internal signal transduction responses to external growth factors.
• The protein is involved in postnatal glycogen storage and energy metabolism.
• Subcellular localization includes both the nucleus and cytoplasm.
