- TWIST2 (also known as Dermo-1, bHLHa39) is a member of the class A basic helix-loop-helix (bHLH) transcription factor family.
- It functions as a transcriptional repressor by binding to E-box consensus sequences (5'-CANNTG-3'), inhibiting the activity of myogenic factors MYOD1, MYOG, MEF2A, and MEF2C.
- TWIST2 plays a key role in repressing pro-inflammatory cytokines such as TNFA and IL1B.
- It inhibits premature or ectopic differentiation of preosteoblast cells during osteogenesis, possibly modulating internal signal transduction responses to external growth factors.
- The protein is involved in postnatal glycogen storage and energy metabolism.
- Subcellular localization includes both the nucleus and cytoplasm.
- In embryonic development, TWIST2 is highly expressed in chondrogenic cells, the undifferentiated mesenchymal layer of skin (dermis), early myeloid cells, and the secretory ependymal epithelium of the choroid plexus primordium. In adults, it is expressed in secreting glandular tissues and tubules.
