Biological Background: TrkB Function and Localization
- Encoded by the NTRK2 gene, TrkB (BDNF/NT-3 growth factors receptor, neurotrophic tyrosine kinase receptor type 2) is a receptor tyrosine kinase that serves as the high-affinity receptor for brain-derived neurotrophic factor (BDNF) and neurotrophin-4 (NTF4), and binds neurotrophin-3 (NTF3) with lower affinity (PubMed:15494731, PubMed:7574684). Alternative splicing generates multiple isoforms, including the full-length kinase-active TrkB, the truncated TrkB-T1, and the Shc-binding TrkB-T-Shc. Related references: PMID:15494731 PMID:7574684
- Upon ligand-induced dimerization and autophosphorylation, TrkB recruits and activates downstream effectors such as SHC1, FRS2, SH2B1, SH2B2, and PLCG1. This triggers the GRB2-Ras-MAPK cascade (neuronal differentiation and neurite outgrowth), the PI3K-AKT1 pathway (cell growth and survival), and PLCG1-PKC signaling (synaptic plasticity).
- Through PLCG1, TrkB also activates NF-kappa-B, promoting transcription of survival genes and suppressing anoikis. It may mediate neurotrophin-dependent calcium signaling in glial cells.
- Subcellular localization includes the cell membrane, endosome/early endosome membranes, axonal and dendritic projections, perinuclear cytoplasm, and postsynaptic density.
- In the CNS, TrkB is expressed in the cerebral cortex, hippocampus, thalamus, choroid plexus, cerebellum, brain stem, and spinal cord. In the PNS, it is found in cranial ganglia, dorsal root ganglia, and sensory nerves. TrkB-T1 is also detected in pancreas, kidney, and heart, while TrkB-T-Shc is predominantly neuronal.

Experimental Guidance and Technical Tips
- The immunogen covers amino acids 32–430 of human TrkB, corresponding to the extracellular domain. This region is expected to recognize all isoforms that share the extracellular region.
- For Western blotting, the predicted molecular weight of full-length TrkB is 92 kDa; however, glycosylation may result in a band shift to a higher apparent molecular weight. A reducing SDS-PAGE and PVDF membrane are recommended.
- In immunofluorescence/ICC and IHC-P, antigen retrieval methods may enhance detection, especially in formalin-fixed paraffin-embedded tissues. Validate the antibody in appropriate positive-control samples (e.g., brain tissue, neuronal cell lines) and include no-primary controls.
- Since this is a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative applications. Consider using it as a capture or detection antibody in ELISA with proper pairing optimization.
