Biological Background: TrkA Function and Localization
- TrkA (high affinity nerve growth factor receptor) is a receptor tyrosine kinase encoded by the NTRK1 gene; it is also known as neurotrophic tyrosine kinase receptor type 1, TRK1-transforming tyrosine kinase protein, and gp140trk. Related references: PMID:1281417 PMID:15488758 PMID:17196528
- It plays a critical role in the development and maturation of the central and peripheral nervous systems by regulating proliferation, differentiation, and survival of sympathetic and sensory neurons through NGF ligand binding and activation (PubMed:1281417, PubMed:15488758, PubMed:17196528). Related references: PMID:1281417 PMID:15488758 PMID:17196528
- Upon NGF binding, TrkA dimerizes, autophosphorylates, and activates downstream signaling cascades including GRB2-Ras-MAPK (differentiation/survival), PLCG1-NF-κB (survival), and Ras-PI3K-AKT1 (survival) (PubMed:1281417). Related references: PMID:1281417
- Alternative splicing produces several isoforms: TrkA-I (non-neuronal), TrkA-II (neuronal), and TrkA-III (neural stem/crest progenitors); TrkA-III lacks NGF binding and has oncogenic activity.
- TrkA localizes to the cell membrane and dynamically traffics through early, late, and recycling endosome membranes upon activation.
- Key structural features include leucine-rich repeats, immunoglobulin domains, a transmembrane helix, and a tyrosine kinase domain; it undergoes phosphorylation and ubiquitination.
- While primarily neurotrophic, TrkA also mediates angiogenesis and can act as an oncogene in tumor settings when aberrantly expressed.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1–280, which includes the extracellular ligand-binding domain; the resulting polyclonal antibody may detect multiple isoforms. Confirm isoform specificity in your cell/tissue type by western blot.
- For western blot, the predicted molecular weight is ~87 kDa (UniProt: 87.5 kDa); note that glycosylation or other modifications may cause band shifts.
- For IHC-P, antigen retrieval conditions may require optimization; validate using known positive control tissues (e.g., brain, dorsal root ganglia).
- As a polyclonal antibody, lot-to-lot variation is possible; pilot testing with your target samples is recommended.
