Biological Background: TrkA Function and Localization

  • TrkA, encoded by the NTRK1 gene, is the high affinity nerve growth factor (NGF) receptor tyrosine kinase. It is also known as neurotrophic tyrosine kinase receptor type 1, TRK1-transforming tyrosine kinase protein, tropomyosin-related kinase A, and p140-TrkA (UniProt: P04629).
  • Upon NGF binding, TrkA homodimerizes and autophosphorylates, triggering downstream signaling cascades, including the Ras-MAPK pathway (cell differentiation and survival), PI3K-AKT pathway (survival), and PLCG1-mediated NF-kappa-B activation (PubMed:1281417, PubMed:15488758, PubMed:17196528).

  • Three major isoforms exist: TrkA-I (widely expressed), TrkA-II (predominantly neuronal), and TrkA-III (pluripotent neural stem/progenitor cells). Isoform TrkA-III lacks the extracellular NGF-binding domain, promoting angiogenesis and oncogenic activity.
  • Subcellularly, TrkA localizes to the cell membrane and is internalized into early, late, and recycling endosomes upon activation.
  • TrkA is critical for nervous system development, and chromosomal rearrangements involving NTRK1 are implicated in certain cancers. It is classified as a proto-oncogene and undergoes alternative splicing, glycosylation, and ubiquitination.

  • The immunogen corresponds to the intracellular region (amino acids 697–796) of human TrkA, which is highly conserved in mouse and rat, supporting observed cross-reactivity.
  • For Western blotting, the predicted molecular weight is approximately 87 kDa. Due to glycosylation, the observed band may appear slightly higher. Reducing conditions and appropriate blocking are recommended.
  • In ELISA applications, consider pairing with a suitable capture antibody and optimizing coating conditions. Validate in relevant biological matrices.
