Biological Background: TRIOBP Function and Localization
- TRIOBP (Tara, TAP68) is an actin-binding protein that directly stabilizes filamentous F-actin and prevents its depolymerization, regulating cell spreading and contraction. Related references: PMID:18194665, PMID:28438837
- It also functions as a linker protein, recruiting factors required for F-actin formation and turnover. Related references: PMID:18194665
- TRIOBP is essential for correct mitotic progression, ensuring proper chromosome segregation. Related references: PMID:22820163, PMID:24692559
- Plays a pivotal role in the formation of stereocilia rootlets in inner ear hair cells.
- Localizes to multiple subcellular compartments: nucleus, cytoplasm, centrosome, midbody, and telomeres.
- Widely expressed with high levels in heart and placenta; also detected in fetal brain, retina, and cochlea.
- Undergoes alternative promoter usage and splicing, producing tissue-specific isoforms.
- Associated with non-syndromic deafness (DFNB28) and is subject to phosphorylation and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 180-310 of human TRIOBP (NP_008963.3). Consider that the polyclonal antibody may recognize multiple isoforms or species orthologs.
- For Western blotting (WB), a predicted band around 261 kDa is expected. Use appropriate high-molecular-weight markers and gradient gels to resolve this large protein. Validate detection in lysates from heart or placenta, which express TRIOBP highly.
- In immunohistochemistry (IHC-P), antigen retrieval may be necessary; optimize using positive control tissues such as retina or cochlea where expression is reported.
- For ELISA, pair with an appropriate coating antigen and validate against recombinant TRIOBP fragments to ensure specificity.
